化潮湿燥方对干燥综合征模型小鼠颌下腺结构和功能的

影响

摘要:干燥综合征是一种常见的自身免疫疾病,其中颌下

腺受到严峻影响。本探究旨在探讨中药方剂“化潮湿燥方”对

干燥综合征模型小鼠颌下腺结构和功能的影响。试验设计为健

康比较组(健康小鼠)、模型组(干燥综合征小鼠)和化潮湿

燥方组(干燥综合征模型小鼠接受化潮湿燥方治疗)。通过观

察颌下腺组织病理学变化、免疫组织化学染色以及检测腺体功

能相关指标,评估化潮湿燥方对于干燥综合征模型小鼠颌下腺

结构和功能的影响。

结果显示,与健康比较组相比,模型组小鼠颌下腺组织呈

现明显的病理学改变,包括腺泡萎缩、上皮细胞变性坏死以及

间质纤维化。免疫组织化学染色结果显示,模型组小鼠颌下腺

组织中细胞凋亡水平明显增加,伴随着缩减的PAS染色阳性细

胞和增加的α-干扰素(IFN-α)表达。此外,模型组小鼠颌

下腺腺液分泌量明显降低。

Abstract: Sjögren's syndrome is a common

autoimmune disease that severely affects salivary

glands, including the submandibular gland. This study

aimed to investigate the effects of the traditional

Chinese medicine formula "Hua Shi Run Zhuang Fang" on

the structure and function of the submandibular gland

in a mouse model of Sjögren's syndrome. The

experimental design included a healthy control group

(healthy mice), a model group (Sjögren's syndrome

mice), and an Hua Shi Run Zhuang Fang group (Sjögren's

syndrome mice treated with Hua Shi Run Zhuang Fang).

The effects of Hua Shi Run Zhuang Fang on the

structure and function of the submandibular gland in

the Sjögren's syndrome model mice were evaluated

through the observation of histopathological changes

in the glandular tissue, immunohistochemical staining,

and measurement of glandular function-related

indicators.

The results showed that compared to the healthy

control group, the submandibular gland tissue of the

model group mice exhibited significant pathological

changes, including atrophy of acini, degeneration and

necrosis of epithelial cells, and interstitial

fibrosis. Immunohistochemical staining results showed

that the level of apoptosis in the submandibular gland

tissue of the model group mice increased significantly,

accompanied by a decreased number of PAS-positive

cells and an increased expression of alpha-interferon

(IFN-α). Additionally, the secretion of submandibular

gland fluid was significantly reduced in the model

group mice.

However, the submandibular gland tissue structure

of the Hua Shi Run Zhuang Fang group mice showed

significant improvement, with restoration of acinar

morphology, reduced epithelial cell death, and less

severe interstitial fibrosis. Moreover, the level of

apoptosis in the submandibular gland tissue of the Hua

Shi Run Zhuang Fang group mice decreased significantly,

the number of PAS-positive cells and IFN-α expression

increased significantly, and submandibular gland fluid

secretion increased.

In conclusion, Hua Shi Run Zhuang Fang has a

significant improvement on the structure and function

of the submandibular gland in a mouse model of

Sjögren's syndrome. It achieves this by reducing

epithelial cell apoptosis and the degree of

interstitial fibrosis, increasing the number of PAS-

positive cells and IFN-α expression, and promoting

submandibular gland fluid secretion. Therefore, Hua

Shi Run Zhuang Fang has potential as a therapeutic

drug for Sjögren's syndrome and deserves further

research and development.
